Establish and maintain a propagation and transformation pipeline for our model amphibious plant Rorippa aquatica (and related species). Assist fundamental, basic tasks for the operation of molecular biology research. Duties include washing and sterilizing laboratory glassware and plasticware (flasks, test tubes, pipettes, etc), making chemical solutions and cell/bacterial culture media, organize chemical inventories, purchase and stock chemical and consumable inventories. Pickup and drop off shipment to/from the laboratory. Assist scientists and students by collecting, organizing, inventorying, and maintaining biological material. Specifically, assist generation, screening, genotyping, harvesting of transgenic plant lines, mutants, and germplasms. Collect, record, organize and verify the accuracy of data, specifically biological materials. Perform calculations, prepare charts, graphs and reports. May use software applications to assemble, manipulate and/or format data. Assist basic molecular biology experiments (e.g. plasmid prep, PCR), basic biochemistry experiments (e.g. recombinant protein expression, purification, SDS-PAGE), and/or basic and advanced microscopy (light microscopy, fluorescent microscopy and confocal laser scanning microscopy). Genome editing using CRISPR/Cas9 constructs. Configure and operate laboratory apparatus and instruments; read and record instrument data; tabulate data; keep laboratory records, and coordinate equipment repairs. Specific equipment may include centrifuges, ultracentrifuge, gel electrophoresis/immunoblot apparatus, autoclaves, dish washer, biosafety cabinet, and freezers/cold chambers. Organize freezers, cold chambers, laboratory stocks of DNA, E.coli, yeast, and plant seeds under the supervision.
